Contractions of human coronary vessels induced by Prostaglandin E2 are mediated via EP3 receptor and modulated by perivascular adipose tissue

چکیده
Introduction: Most of the blood vessels are surrounded by perivascular adipose tissue (PVAT) which could regulate vascular functions. Prostaglandin E2 (PGE2) released by PVAT could regulate vascular tone depending on which PGE2 receptors (EP) are expressed by the vessel (vasocontriction via EP1 / EP3, vasodilatation via EP2 / EP4 receptors). During vascular pathologies associated with inflammation such as atherosclerosis, the release of PGE2 is increased. The aim of this study is to determine the effects of PVAT and PGE2 on vascular tone of human coronary vessels (HCV), with or without atherosclerosis).

Importance of adipocyte cyclooxygenase-2 and prostaglandin E2-prostaglandin E receptor 3 signaling in the development of obesity-induced adipose tissue inflammation and insulin resistance.
We examined the involvement of adipocyte cyclooxygenase-2 (COX-2) and prostaglandin E2 (PGE2)-prostaglandin E receptor (EP)3-mediated signaling during hypertrophy and hypoxia in the development of obesity-associated adipose tissue (AT) inflammation and insulin resistance. The experiments were conducted with high-fat diet (HFD)-induced obese rats, db/db mice, human subjects, and 3T3-L1 and the h...
متن کاملRegulation of Vascular Smooth Muscle Tone by Adipose-Derived Contracting Factor
Obesity and arterial hypertension, important risk factors for atherosclerosis and coronary artery disease, are characterized by an increase in vascular tone. While obesity is known to augment vasoconstrictor prostanoid activity in endothelial cells, less is known about factors released from fat tissue surrounding arteries (perivascular adipose). Using lean controls and mice with either monogeni...
متن کاملProstaglandin E2 in Soluble Factor-Mediated Immune Suppression by First Trimester Decidual Cells
Several studies have shown that decidual tissue produces soluble factors which can mediate a variety of immunosuppressive activities. In order to assess whether prostaglandin E2 (PGE2) was present in supernatants produced by explant culture of 10 human specimens of first trimester decidua. The effect of the supernatants from the two different series (with and without indomethacin treatment) ...
متن کاملImmunolocalization of the four prostaglandin E2 receptor proteins EP1, EP2, EP3, and EP4 in human kidney.
Four prostaglandin E2 receptor subtypes designated EP1, EP2, EP3, and EP4 have been shown to mediate a variety of effects of prostaglandin E2 (PGE2) on glomerular hemodynamics, tubular salt and water reabsorption, and on blood vessels in the human kidney. Despite the important role of renal PGE2, the localization of PGE2 receptor proteins in the human kidney is unknown. The present study used a...
متن کاملInteractions between vascular wall and perivascular adipose tissue reveal novel roles for adiponectin in the regulation of endothelial nitric oxide synthase function in human vessels.
BACKGROUND Adiponectin is an adipokine with potentially important roles in human cardiovascular disease states. We studied the role of adiponectin in the cross-talk between adipose tissue and vascular redox state in patients with atherosclerosis. METHODS AND RESULTS The study included 677 patients undergoing coronary artery bypass graft surgery. Endothelial function was evaluated by flow-medi...
